摘要:
以水稻两优培九剑叶为材料,通过蓝绿温和胶电泳对其类囊体膜蛋白复合物进行分析。亚细胞组分是植物蛋白质组学研究的重点,而高度疏水且结构复杂的膜蛋白复合物在传统二维电泳中难以得到很好分离,故采用纯化完整叶绿体,温和去污剂增溶膜蛋白,非变性梯度凝胶电泳的方法,直观地分离并展示了两优培九水稻剑叶类囊体9条蛋白质复合物条带,并显示出丰度与组成的差异。以蓝绿温和胶为一向,含尿素的变性电泳为二向的二维电泳将两优培九水稻剑叶类囊体膜蛋白复合物分离为亚基,得到51个蛋白质点,为对水稻光合机构响应逆境等变化的蛋白组学研究提供方法。
