摘要:
为探讨黑素皮质素受体4(MC4R)基因多态性对山羊体质量的影响,以波尔山羊为研究对象,采用池DNA为模板,PCR扩增MC4R基因,Sanger测序法筛选其在波尔山羊中的SNPs位点,PCR-RFLP方法进行多态位点分型,单因素方差分析不同基因型间波尔山羊各月龄体质量的差异。结果表明,编码区502 bp处新检测到A/G单碱基颠换,位于BseJⅠ酶切位点内,在波尔山羊群体中存在AA(0.779 1)、AG(0.202 5)和GG(0.018 4)3种基因型,关联分析发现AA基因型个体初生体质量、1周龄体质量显著高于AG基因型(P<0.05),1周龄体质量极显著高于GG基因型(P<0.01),2周龄、2月龄、3月龄体质量显著高于GG型(P<0.05)。连锁分析发现,A502G、C693T、A946C和C986T位点共存在8种不同的基因型组合,AACCAACC型个体初生体质量显著高于AGCCAACC型(P<0.05),AACCAACT型个体周岁体质量显著高于AACTACCC型和AGCTACCC型(P<0.05)。综上所述,MC4R基因的A502G位点A基因对山羊早期体质量增加更有利,基因型组合AACCAACT对山羊周岁体质量增加更有利,可以作为山羊生长性状选择的候选分子标记。
