摘要:
为建立一种准确和快速鉴定斑点叉尾鱼回遗传性别方法,根据其性别连锁SNP(single nucleotide polymorphism)位点,设计外围引物、SNP特异性延伸引物和DNA探针。性别连锁SNP特异性引物延伸反应结束后,将扩增终产物与DNA探针杂交,并应用基于免疫胶体金技术的核酸检测试纸检测性别连锁SNP,从而实现遗传性别分子水平的可视化鉴定。结果显示,该方法能够准确地鉴定斑点叉尾鱼回的遗传性别,与基于性别连锁微卫星标记的遗传性别鉴定方法所得结果一致,表明斑点叉尾鱼回性别连锁SNP特异性引物延伸反应联合基于免疫胶体金技术的核酸试纸能够准确、快速地完成斑点叉尾鱼回的遗传性别鉴定。
