摘要:
类胡萝卜素各组分合成的调节和积累对辣椒果实颜色形成具有重要的作用,胡萝卜素羟化酶(CRTZ)是植物类胡萝卜素合成代谢途径中的关键酶,对果色形成具有重要影响,探究了CRTZ基因调控辣椒果色机制,为选育高类胡萝卜素辣椒新品种提供理论依据。以红色辣椒PC01、SP01及其橙色、黄色突变体PC02、SP02以及观赏椒Z1为材料,分析不同果实颜色辣椒中CRTZ基因的序列差异和表达特性。研究结果表明,5个品种辣椒叶片组织中均能克隆到CRTZ全长基因,且序列无差异;CRTZ基因其CDS全长为510 bp,编码315个氨基酸。序列比对和进化树分析表明CRTZ蛋白与番茄、彭氏茄具有较高同源性。实时荧光定量PCR(qRT-PCR)分析结果表明:随着植株发育CRTZ基因在5个辣椒品种中表达整体呈上升趋势,且主要在茎、花、果实中表达,在橙色突变体PC02的表达量总体高于其在野生型PC01中的表达量,而在黄色突变体SP02中,其表达量低于野生型SP01。本试验在辣椒叶片组织中都克隆到全长CRTZ基因,所以导致这种现象不是由于结构变异所致,然而在各组织中该基因的表达量不同,说明基因的表达量高低可能对辣椒果色调控有着重要影响。
