摘要:
采用DNA条形码序列对不同品种的金银花进行鉴定,为金银花的鉴定提供分子依据。主要选择ITS2和 psbA-trnH 序列进行评价,以扩增及测序成功率、变异位点数和K-2-P距离等指标评价各序列的鉴定能力。此外,基于MEGA 4.0分析不同品种金银花序列种间K-2-P遗传距离并构建NJ树。结果表明:不同品种金银花ITS2和 psbA-trnH 序列的扩增及测序成功率均较高,扩增成功率为100%,ITS2和psbA-trnH的测序成功率分别为72.7%、91%,且两者均存在较多的变异位点,可以有效地区分金银花不同品种。说明ITS2和psbA-trnH可以作为鉴定金银花不同品种的优势条形码组合。
