摘要:
为了从自身的角度提高绵羊对传染性胸膜肺炎的抗感染能力,采用MSAP(甲基敏感扩增多态性)的方法对20只患病绵羊和20只正常绵羊基因组DNA甲基化与传染性胸膜肺炎关联性进行了分析。以绵羊在患传染性胸膜肺炎前后的全基因组DNA为研究对象,经过同裂酶HpaⅡ和MspⅠ对其进行双酶切,最后通过测序胶的聚丙烯凝胶电泳处理从16对MSAP选择性引物组合中挑选出4对引物的扩增结果,进行绵羊患病前后甲基化变化的评估。分析结果,传染性胸膜肺炎的患病与否,与绵羊的甲基化程度有一定的关联,通过统计学分析得出患病绵羊甲基化水平普遍高于非患病绵羊,同时甲基化多态性也随之升高。结果对研究动物疾病与动物自身表观遗传之间关联起到一定的参考意义,也为今后的研究提供了理论依据。
