摘要:
以连香树(Cercidiphyllum japonicum)下胚轴为外植体进行离体培养,以MS、1/2MS和B5为基本培养基,附加不同浓度的细胞分裂素(CPPU、6-BA)及生长素(2,4-D)诱导下胚轴直接再生不定芽。结果表明,不定芽未经过愈伤组织而直接产生于下胚轴的表皮或近表皮等表层细胞;不同激素浓度和组合、不同培养基对不定芽的分化有影响;下胚轴的不同部位不定芽的分化能力差异显著。适宜不定芽分化的最佳培养体系为1/2MS+AgNO3 3.5 mg/L+蔗糖3.5%+琼脂0.5%+2,4-D 0.4 mg/L+ CPPU 1.5 mg/L,最佳外植体为靠近子叶端的下胚轴部分,分化率最高达89.3%,最高的平均每个外植体再生芽数达到18.07;生根培养基选用1/4MS+蔗糖4.5% +琼脂0.5%+活性炭2.5%+ DA-6 3.5 mg/L+CSN 2 mg/L,生根率为83.6%,平均根数8.2。
