摘要:
采用RT-PCR方法对四川绵阳地区采集的疑似猪繁殖与呼吸综合征感染病料进行克隆,并对ORF5基因进行测序及遗传进化分析。Nsp2基因扩增结果表明,获得的15个猪繁殖与呼吸综合征阳性毒株均为Nsp2基因存在缺失的变异株。ORF5基因测序结果表明,15株ORF5基因均由603 bp编码、200 个氨基酸组成,各毒株之间的氨基酸同源性为87.6%~100.0%,与GenBank 中13 株参考毒株的序列同源性在57.2%~99.0%之间,与HUN4、JXA1、TJM、GXHZ12等HP-PRRSV 代表株ORF5的相似性高达90%~99%。推导氨基酸分析结果表明,各毒株的ORF5氨基酸序列因各场的不同而差异显著,存在一些突变位点,同时同一场内的不同毒株间的差异也较大,多数毒株在信号肽、跨膜功能区都存在突变位点,各毒株的潜在糖基化位点也存在差异;遗传进化树分析结果表明,15 株ORF5 序列均属于北美株,大部分毒株序列与2006年暴发的JXA1高度同源。总体上,各毒株之间没有呈现明显的地域性。
