摘要:
为了观察秋水仙胺(demecolcine,DEME)诱导牛卵母细胞的显核效果,从吉林省长春市某屠宰场收集牛卵巢后采用抽吸法获取卵泡液,在体视显微镜下选择卵丘完整、胞质均匀的卵丘-卵母细胞复合体(COCs),微滴法体外成熟(in vitro mature,IVM)培养19~22 h,在0.3%透明质酸酶中旋涡振荡去除卵母细胞周围的卵丘细胞,挑选排出第一极体的卵母细胞随机分组,分别进行DEME浓度和作用时间诱导牛卵母细胞显核的研究。结果表明,DEME处理60、90、120 min组间显核率差异不显著(P>0.05),但是60、120 min组的显核率显著高于DEME处理 30 min 组(P<0.05)。当DEME浓度为0.5 μg/mL时,显核率最高,为78.9%,显著高于0.3、0.4 μg/mL组(P<0.05),但是与 0.6 μg/mL 组差异不显著(P>0.05)。牛的COCs经IVM培养19、20、21、22 h,结果发现培养22 h组显核率最高,显著高于培养19 h组(P<0.05),但是与培养20、21 h组差异不显著(P>0.05)。可见牛卵母细胞经IVM培养 20~22 h后,用0.5 μg/mL DEME处理60 min可以有效诱导其显核。
