摘要:
以芒果基因组DNA为模板,采用正交试验对模板DNA含量、引物浓度、2×Taq PCR Master Mix含量进行3因素5水平正交试验优化。结果表明,相关序列扩增多态性PCR(sequence related amplified polymorphism,SRAP-PCR)最佳反应体系为:30 ng 模板DNA、0.3 μmol/L 引物、10 μL 2×Taq PCR Master Mix,总体积为25 μL。运用该体系对10份芒果种质材料进行验证,证明该体系稳定可靠,并从100对引物组合中筛选出扩增条带清晰、多态性丰富的36对引物组合。
