摘要:
抑制素(INH)是性腺组织分泌的一种糖蛋白,可通过抑制促卵泡素分泌和性腺组织发育,在动物繁殖活动中起着重要的调控作用。通过对INH α亚基免疫,中和动物体内的INH,可解除INH对动物繁殖活动的抑制作用,从而提高动物的繁殖力。为获得大量重组的INH α亚基蛋白,满足生产的需要,本研究对发酵罐高密度发酵工艺进行探索,并分别对发酵培养基的类型、乳糖诱导浓度、诱导时间进行优化。结果表明,采用TB培养基发酵至20 h,菌体D600 nm值达到最大,为22.84,菌体总湿质量为25.48 g/L,显著高于其他2种培养基,综合比较细菌生长趋势、菌体总湿质量以及培养基成本等因素,TB培养基较适用于重组INH的发酵罐生产;终浓度为10 g/L的乳糖诱导5 h可达到最高蛋白表达量,占菌体总蛋白的50%以上,发酵结束时D600 nm值达到36,细菌总湿质量达到28.66 g/L。
