摘要:
利用精密肝切片培养技术研究2,3,7,8-四氯二苯并-p-二恶英(TCDD)对建鲤肝组织的损伤作用,将精密肝切片组织分为5个处理组,每个处理组4个重复,将不同浓度TCDD(浓度设定为0.00、0.05、0.10、 0.30、 0.60 μg/L)处理肝切片3 h后,收集上清培养液及肝切片组织测定丙氨酸转氨酶(GPT)、天冬氨酸转氨酶(GOT)、乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)、丙二醛(MDA)、三磷酸腺苷(ATP)等生化指标含量变化。结果表明:TCDD浓度在 0.30 μg/L时,对精密肝切片组织损伤较大,可以显著提高上清培养液中GPT、GOT、MDA的含量,显著提高切片匀浆上清液中LDH的活性值,与空白对照组相比,差异极显著(P<0.01);可以显著降低切片匀浆上清液中ATP、GSH-PX的含量,与空白对照组相比,差异极显著(P<0.01);显著降低上清培养液中SOD活性值,与空白对照组相比,差异显著(P<0.05)。这说明采用TCDD进行急性染毒后,可以造成建鲤肝组织的损伤,使机体组织处于氧化应激状态,产生一系列氧化应激反应。
