摘要:
以梅花的花朵为材料,采用RT-PCR与RACE相结合的方法,从“三轮玉蝶”梅花的花朵中克隆到1个全长1 322 bp的氧-甲基转移酶cDNA,命名为PmOMT,该基因编码377个氨基酸的开放性阅读框,具有植物氧-甲基转移酶基因典型的3个保守基序,与月季(R. chinensis var. spontanea)的甲基(异)丁香酚转移酶基因RcOMT1具有较高的同源性。构建系统发育进化树结果表明,PmOMT与RcOMT1亲缘关系最近,同属COMTs类基因。荧光定量PCR分析发现,PmOMT基因的相对表达量与甲基丁香酚色谱峰面积的变化趋势相似,推测该基因可能参与甲基丁子香酚的生物合成。
