摘要:
建立采用高效液相色谱-串联质谱(high performance liquid chromatography-tandem mass spectrometry,HPLC-MS/MS)测定绵羊血浆中催产素(oxytocin,OT)的方法。经乙腈沉降法提取血浆样品,在多反应离子监测(multiple reaction monitoring,MRM)模式下进行检测,标准加入法定量。通过该方法获得标准曲线线性方程为y=1.448x+544.400,相关系数为0.998,线性范围在50~10 000 pg/mL,方法检出限为10 pg/mL,定量限为50 pg/mL。应用 HPLC-MS/MS法测定醋酸去甲雄三烯醇酮-17β雌二醇(trenbolone acetate and estradiol-17β,TBA-E2)联合处理阉公羊血浆中OT浓度变化,其中对照组催产素浓度为55~80 pg/mL,TBA-E2处理组为75~160 pg/mL,表明TBA-E2可提高阉公羊血浆中的OT浓度。该方法简便、快速、准确,可用于血浆样品中OT的检测。
