摘要:
由己烯雌酚(diethylstilbestrol,DES)经过一系列化学反应,合成含有1个羟基活性基团的半抗原己烯雌酚-单羧基丙基醚(DES-MCPE);应用活泼酯法,使DES-MCPE与牛血清白蛋白(BSA)、卵清蛋白(OVA)偶联,合成人工免疫抗原DES-BSA和包被抗原DES-OVA,采用紫外分光光度法(UV)和聚丙烯酰胺凝胶电泳(SDS-PAGE)进行鉴定,初步判定偶联成功。用DES-BSA以60 μg/只的剂量免疫3只BALB/c小鼠,4次免疫后,采集血清,使用间接ELISA和间接竞争ELISA分别测定抗血清的效价与抑制,3只小鼠的效价均可达到1×104以上,其中2号小鼠的半抑制率(IC50)为18.221 μg/L,表明已获得灵敏、特异、高效价的抗血清。
