摘要:
为明确生防细菌 G10菌株在芝麻立枯病生物防治中对环境条件的耐受性,首先利用平板培养法检测不同浓度条件下G10菌株代谢产物对立枯丝核菌的抑制作用,再利用对峙培养法分别测定pH值、温度、盐等因素对菌株代谢产物抑菌效果的影响。同时,对生防菌的定殖能力及其生物膜的形成也进行测定。结果表明,随着G10菌株发酵液浓度的提高,抑菌率逐渐提高,不同浓度的发酵液抑菌率差异显著(P<0.05);对试验数据进行线性回归分析,估算出G10菌株代谢产物对立枯丝核菌的EC50为12.68%;G10菌株代谢产物在pH值为6~9范围内稳定,可发挥正常的抑菌作用;在pH值≤5及pH值≥10条件下,其代谢产物的抑菌率逐渐下降。该菌代谢产物在40~50 ℃温度范围内抑菌率差异不显著,当温度≥60 ℃时代谢产物抑菌率显著下降(P<0.05);几种盐离子对代谢产物抑菌率有一定的负作用;紫外线照射10 min内该菌代谢产物抑菌性能稳定,紫外线照射时间≥15 min时抑菌率逐渐下降。G10菌株在LB培养基和Landy培养基上均具有形成生物膜的能力,利用LB培养基培养时,该菌株生长形成的生物膜D570 nm平均值为1.125,利用Landy培养基培养时,形成生物膜的D570 nm平均值为1.632。以上结果显示,该菌代谢产物对温度和pH值稳定范围宽,耐受紫外线能力强,且菌株具有较强的形成生物膜的能力。研究结果可以为G10菌株生防制剂的开发提供理论依据。
