摘要:
为了根据猪弓形虫感染血清差异表达多肽,建立一种新的质谱检测方法,采集江苏泰州地区部分养殖场育肥猪血清,利用间接血凝法(IHA)分析弓形虫感染状况,将IHA阳性与阴性血清分别归为感染组和对照组,采用磁珠纯化试剂盒富集血清多肽,应用基质辅助激光解析电离飞行时间质谱(MALDI-TOF Mass Spectrometry)检测,并结合flexAnalysis 2.0与Clin ProTools软件分析检测图谱并建立检测模型。对海陵、高港、姜堰部分猪场的IHA检测显示,猪弓形虫感染率分别为16%(8/50)、16%(8/50)、18%(9/50)。质谱检测m/z 800~12 000区段发现,弓形虫IHA阳性与阴性相比较,在育肥猪血清中共有9个多肽峰表达有显著性差异,其中m/z 1 608.59、1 971.84、5 004.01、4 617.49、7 808.23的5个多肽峰的表达显著上调,而m/z 1 233.01、8 007.66、2 356.37、3 735.65的4个峰呈显著下调,应用ClinProTools建模方程建立弓形虫感染猪的检测模型。可见MALDI-TOF质谱结合ClinProTools分析可建立猪弓形虫感染检测新方法,为弓形虫的诊断提供了一种新思路。
