摘要:
拟通过高效液相色谱研究时间、温度、pH值、氯化钠和次氯酸钠对肌红蛋白血红素辅基稳定性的影响。设肌红蛋白在4 ℃下放置的时间为0、1、3、5、7 d;处理温度依次为4、20、30、40、50 ℃;缓冲液pH值依次为3、5、7、9、11;氯化钠质量分数依次为1%、2%、3%、4%、5%;次氯酸钠处理时间依次为1、2、3、4、5、6 h。结果表明,随着时间的延长,血红素辅基损失逐渐增大;温度和盐度对血红素辅基结构稳定性的影响不是很明显;血红素辅基在偏碱性环境下较稳定,而在酸性环境中血红素辅基损失速率明显提高;次氯酸钠会破坏血红素辅基的稳定性,并且随着处理时间的延长,这种作用逐渐增强。
