摘要:
利用分子克隆技术和生物信息学方法,对獭兔CREB1基因编码区序列(coding sequence,CDS)进行克隆和生物信息学特征分析,同时比较其在不同毛色獭兔的皮肤组织中的表达水平差异。结果表明,CREB1基因CDS长 984 bp,共编码327个氨基酸,在不同的哺乳动物中具有较高的同源性,系统进化情况与其亲缘关系远近一致。经Hopfield、Swiss-model等预测发现,CREB1基因编码蛋白为亲水蛋白质,其二级结构包含α螺旋(h)33.33%、无规则卷曲(c)44.95%、延伸链(e)21.71%;三级结构为1条简单的螺旋链。荧光定量PCR结果显示,CREB1基因在黑色獭兔和青紫蓝色獭兔皮肤组织中的表达量极显著高于白色獭兔(P<0.01),且在黑色獭兔中表达量最高。该结果为进一步探索CREB1基因在哺乳动物毛色形成过程中的功能奠定了基础。
