摘要:
以进境的菠菜种子为试验材料,采用特异性引物逆转录PCR(RT-PCR)检测鉴定菠菜潜隐病毒(SpLV),并对扩增得到的特异性片段进行进一步测序验证。结果表明,采用的3对引物分别扩增得到了3种不同大小的特异性片段,其中引物对SpLV-RNA2-F/R扩增的RNA2组分产物量最大,条带清晰,最适合日常SpLV的检测鉴定,目的条带测序结果与GenBank发布的菠菜潜隐病毒分离物的同源性为97.87%~100.00%,进一步说明进境菠菜种子感染了菠菜潜隐病毒。这是我国首次从进境菠菜种子上截获菠菜潜隐病毒。
