摘要:
为了建立籼稻Kra Dang Ngah的再生体系,以其成熟胚作为试验材料,研究不同种类植物激素及其浓度配比对诱导愈伤率、愈伤组织形态学特征、分化及植株再生的影响。结果表明:2,4-D可诱导愈伤组织产生,但愈伤组织诱导率极低;2,4-D和其他植物激素配合使用显著提高了诱导率,在2 mg/L 2,4-D、1 mg/L NAA、1.0 mg/L 6-BA 和 0.5 mg/L KN 的条件下可获得较高的愈伤组织诱导率,为56.3%。继代培养基中添加激素浓度为诱导培养基中的一半可获得较好的增生的胚性愈伤组织,形态学特征为淡黄色,比较致密、较硬、呈颗粒状结构,没有褐化出现,表现出很强的分化能力。再生培养中,MS培养基中添加0.5 mg/L NAA、1.0 mg/L 6-BA 和2 mg/L KN可获得较高的分化率(达到75.5%)和再生率(33.3%),表明6-BA和Kn的配合使用显著提高了分化率。
