摘要:
为明确实验室内饲养的3个灰飞虱地理种群(云南楚雄、江苏南京、江苏建湖)共生菌Wolbachia的感染情况以及Wolbachia对灰飞虱生殖的影响,首先利用PCR技术对这些种群的Wolbachia进行检测,发现所有种群均感染了Wolbachia,楚雄、南京、建湖种群的感染率分别为91.5%、66.1%、84.0%。接着用Wolbachiawsp基因的特异性引物从3个地理种群中都扩增出1段599 bp的基因片段,通过测序发现这3个种群的Wolbachia具有完全相同的wsp基因序列,同属于Con株系。最后,在严格的试验条件下对建湖种群的灰飞虱进行了杂交试验,结果表明:Wolbachia对灰飞虱生殖表现出高强度的单向胞质不亲和;Wolbachia可能对宿主生殖具有负面影响;另外,研究还发现亲本双方全部感染Wolbachia时,其子代感染率仅为82.14%,Wolbachia的卵传率未达到100%,这可能与Wolbachia在灰飞虱种内的垂直传播有关。
