摘要:
60尾鲫鱼单剂量(12 mg/kg)口服给予阿苯达唑后采集血样,进行阿苯达唑及其亚砜在鲫鱼体内的药动学研究;40尾鲫鱼以同等剂量连续口服给药5 d,之后采集肌肉样品,检测药物浓度,分析残留消除的过程;利用反相高效液相色谱法(RP-HPLC)检测血浆和肌肉中阿苯达唑以及其代谢产物阿苯达唑亚砜的浓度,药物浓度-时间数据采用3P97药动学程序软件处理。结果表明,鱼口服阿苯达唑后,血浆中阿苯达唑亚砜药物浓度-时间数据符合一级吸收一室开放模型,t1/2Ka为10.91 h,t1/2Ke为11.95 h,tpeak为16.46 h,Ka为0.06 h-1Ke为0.06 h-1Cmax为0.28 mg/L,AUC为12.56 mg·h/L,药动学参数表明阿苯达唑亚砜在15~18 ℃水温下,在鲫鱼体内吸收缓慢,滞留时间和半衰期较长,分布容积较小,血药浓度低。在20 ℃水温下,多剂量口服阿苯达唑残留消除试验表明,停药后4 d肌肉中阿苯达唑亚砜的浓度为0.787 mg/kg,停药后8 d为0.064 mg/kg,停药后24 d药物浓度低于检测限。
