摘要:
拟克隆野葛钙离子依赖性核酸酶PlCaN基因全长cDNA序列,并对其进行生物信息学分析,同时构建其原核表达载体。提取野葛材料总RNA作为模板,利用逆转录PCR(RT-PCR)、 cDNA末端快速扩增技术(rapid-amplification of cDNA ends,简称RACE)等方法获取野葛PlCaN的cDNA全长序列,利用相关软件和在线网站进行生物信息学分析,构建原核表达载体pBRT7-PlCaN并鉴定。结果表明,PlCaN的cDNA全长1 008 bp,编码335个氨基酸,预测蛋白质相对分子质量为37.3 ku,理论等电点(pI)为9.4,为亲水性蛋白,无信号肽,表明原核表达载体pBRT7-PlCaN构建成功,为探究其在不同组织中的表达情况,建立原核表达体系,并为后期PlCaN的功能研究提供理论基础。
