摘要:
利用RT-PCR以及ELISA对采自云南省昆明市的君子兰样品进行鉴定,确定了其病原物为朱顶红褪绿环斑病毒(hippeastrum chlorotic ringspot virus,HCRV)。这是首次在君子兰(Clivia miniata)中检测到番茄斑萎病毒属(Tospovirus)病毒,为明确其序列特征,设计了HCRV S RNA全序列扩增引物,进一步克隆了该分离物的S RNA全基因组序列,并进行了序列分析。结果表明,HCRV-CM分离物S RNA全长2 749 nt(GenBank登录号:KY484836),与已报道的HCRV基因结构一致。基于核壳体蛋白N基因序列的系统进化分析显示HCRV-CM与来自云南的HCRV株系聚为一支,说明HCRV-CM与HCRV云南分离物亲缘关系较近。
