摘要:
利用线粒体细胞色素b(cytochrome b,简称Cytb)基因和控制区(D-loop)序列作为分子标记,调查洪泽湖湖鲚的遗传多样性。结果表明,Cytb基因和D-loop区序列碱基A+T含量均高于G+C含量,显示碱基组成具有偏倚性。40条Cytb基因序列检出14个变异位点,定义13个单倍型,单倍型多样性和核苷酸多样性分别为 0.762±0.067、0.0018 2±0.000 28,平均碱基差异数为2.073;40条D-loop区序列检出54个变异位点,定义17个单倍型,单倍型多样性和核苷酸多样性分别为0.727±0.078、0.006 10±0.001 00,平均碱基差异数为7.378。13个Cytb基因单倍型之间的遗传距离在0.001~0.004之间,17个D-loop区单倍型之间的遗传距离在0.001~0.018之间,邻接法(neighbour-joining method,简称NJ)系统进化树显示单倍型聚为1支。Cytb基因和D-loop区序列中性检测Tajimas D和Fus Fs结果产生的DFs值为负值,且Cytb基因的Fs值统计结果有极显著差异(P<0.01)。同时,Cytb基因序列的岐点分布图呈现单峰型,均表明洪泽湖湖鲚近期经历了种群扩张。整体来看,洪泽湖湖鲚资源具有较高的遗传多样性,呈现出高单倍型多样性和低核苷酸多样性的特征。
