摘要:
旨在阐明污染小麦的真菌的结构组成并分析污染小麦的真菌与麦粒中呕吐毒素(DON)浓度之间的关系。采用高效液相色谱法(HPLC)测定麦粒中的DON含量,选取DON浓度为高、中、低[DON含量分别为(2 982±1 534)、(972±251)、(42±1) μg/kg]的小麦样品各12份,抽提3组小麦微生物DNA进行真菌测序。结果表明,通过基因组DNA提取、高通量测序、生物信息学分析后,共得到820个运算分类单元(operational taxonomic unit,简称OTU)。基于不同分类水平的分析可知,其优势菌门为子囊菌门(Ascomycota)、担子菌门(Basidiomycota),平均相对丰度分别为46964 0%、0.722 1%;优势菌属为链格孢属(Alternaria)、附球菌属(Epicoccum),分别占16.987 9%、5.570 1%。3组小麦中的真菌物种数量有显著差异(P<0.05)。由研究结果可知,小麦中存在一定的病原真菌污染,如链格孢属(Alternaria)、葡萄孢属(Botrytis),有必要对携带真菌毒素DON小麦中的真菌结构进行检测,并分析两者间的关系。
