摘要:
稻草和牛粪堆制发酵是生产双孢蘑菇的重要环节,分析发酵过程中微生物群落动态变化有助于揭示培养料发酵的机理以及探讨其与温室气体排放之间的关系。以稻草和牛粪为原料,采用磷脂脂肪酸(PLFA)生物标记法分析不同碳氮比(C/N=28、33、38、43)条件下发酵料在不同发酵阶段(第1次高温期、第2次高温期、一次发酵结束、二次发酵结束)的微生物群落变化。结果表明,发酵过程中共分离到24种碳链长度在12~24的PLFA;随发酵进程加深,不同C/N处理的微生物PLFA种类和丰度都有变化,但丰度最高的9个生物标记均为i16 ∶ 0和a17 ∶ 0[革兰氏阳性菌,Gram(+)],15 ∶ 0和18 ∶ 0(细菌),18 ∶ 2ω6t,9t和18 ∶ 1ω9c(真菌),16 ∶ 1ω7c、16 ∶ 1ω9c和cy19 ∶ 0[革兰氏阴性菌,Gram(-)]。不同C/N处理的特征脂肪酸含量在发酵过程中均呈现出细菌>Gram(+)>真菌、Gram(-)>放线菌,并且总PLFA、细菌和Gram(+)PLFA含量均呈现出先降低后增加的趋势,然而Gram(-)和放线菌呈现出逐渐增加的趋势。在第1次和第2次高温期,随着C/N增加,总PLFA、细菌和Gram(+)PLFA含量均有增加的趋势。二次发酵结束时,C/N=33处理的总PLFA、细菌、Gram(-)、放线菌PLFA含量显著高于其他处理,C/N=38处理次之。相关性分析表明,放线菌与发酵温度、N2O排放速率均呈显著负相关。
