摘要:
为了研究日粮中添加辣椒碱对脂多糖应激仔猪急性肝损伤的作用机制,选取初始体重相近的28日龄断奶仔猪24头,随机分为3组,即对照组、脂多糖组饲喂基础日粮,辣椒碱组在基础日粮中添加800 mg/kg的辣椒碱,试验周期35 d,脂多糖组和辣椒碱组仔猪均于采样前4 h按照100 μg/kg的剂量腹腔注射脂多糖,对照组仔猪腹腔注射等剂量的生理盐水。结果显示:与对照组相比,脂多糖组血清中谷草转氨酶、谷丙转氨酶、碱性磷酸酶活性显著升高(P<0.05),肝脏还原型谷胱甘肽含量、谷胱甘肽过氧化物酶活性、总抗氧化能力显著降低(P<0.05),丙二醛、蛋白羰基含量显著上升(P<0.05);与脂多糖组相比,辣椒碱组能显著降低血清谷草转氨酶、碱性磷酸酶酶活性(P<005),显著提高肝脏还原型谷胱甘肽含量、谷胱甘肽过氧化物酶活性、总超氧化物歧化酶活性、总抗氧化能力水平(P<005),显著降低蛋白羰基及8-羟基脱氧鸟苷含量(P<0.05)。与对照组相比,脂多糖组仔猪肝脏PI3K、CAT、GPX1、SOD1表达下调(P<0.05),SOD2表达上调(P<0.05),LC3B、PCK1、FUNDC1表达上调(P<0.05),BNIP3L表达下调(P<0.05);与脂多糖组相比,辣椒碱组会显著上调肝脏中PI3K、Nrf2、CAT、GPX1、NQO1、SOD2表达(P<005);LC3B、P62、PCK1、Drp1、Mfn1、BNIP3、FUNDC1表达显著下调(P<0.05)。辣椒碱能够增加肝脏抗氧化酶活性,降低氧化代谢产物水平,通过调控PI3K/AKT/Nrf2信号通路改善肝脏抗氧化能力,维持肝脏线粒体功能稳定,从而缓解脂多糖诱导的断奶仔猪急性肝损伤。
