摘要:
为探讨不同浓度的茉莉酸甲酯(MeJA)对雪茄烟叶发酵过程中细菌群落结构的影响,以期为优化雪茄发酵工艺提供科学依据。MeJA作为一种重要的植物次级代谢物,在植物信号传导和环境胁迫应答中发挥关键作用,但其在雪茄发酵中的作用及其对菌群结构的影响尚未得到系统研究。采用茄衣海研101品种的雪茄烟叶,设置不同浓度的MeJA处理(0、10、50、100、200、500 μmol/L),每个处理重复3次。在发酵前对烟叶进行喷施处理,并分别在喷施后(0 d)及发酵30 d后取样。通过Illumina NovaSeq平台进行16S rRNA高通量测序,分析细菌群落的物种注释及丰度,进行α多样性、β多样性分析,并利用OTU聚类和LEfSe分析群落结构。结果表明,葡萄球菌属(Staphylococcus)为所有处理中的优势菌属,且在100 μmol/L MeJA(C4)处理的丰度最高。假单胞菌属(Pseudomonas)和迪茨氏菌属(Dietzia)为添加MeJA后的优势菌属,其丰度较对照组明显提升。不同浓度的MeJA明显影响了雪茄发酵过程中的细菌多样性和群落组成,尤其是100 μmol/L MeJA处理对细菌多样性抑制最为明显。在纲水平上,γ-变形菌纲和芽孢杆菌纲在所有处理处理的相对丰度最高,表明其在雪茄发酵中可能发挥核心作用。在属水平上,MeJA处理明显改变了细菌群落结构,其中低浓度MeJA(10、50 μmol/L)促进了乳酸菌和拟杆菌等代谢功能菌群的增长,而高浓度MeJA(100、200 μmol/L)则增加了梭菌属和放线菌的丰度。由此可知,MeJA浓度对雪茄发酵中的细菌群落结构具有明显的调控作用。不同浓度的MeJA通过改变细菌群落的多样性和组成,影响雪茄发酵过程中的代谢途径和风味品质。本研究为利用外源添加剂MeJA精准调控雪茄发酵菌群提供了理论支持,也为进一步优化雪茄发酵工艺和提升产品品质提供了新思路。
