摘要:
为探究IMPAD1基因在鸡肌肉发育和脂肪沉积中的作用,以S3、H和F系鸡为试验素材,利用荧光定量PCR检测IMPAD1在鸡不同发育时期不同组织及细胞中的表达,并做相关性分析。结果表明:在腹部脂肪组织中,IMPAD1在2、15周龄H系和F系鸡中的表达量极显著高于S3系(P<0.01);在皮下脂肪组织中,2、5周龄时F系鸡极显著高于其他品系(P<0.01);在胸肌组织中,2、5、10、15周龄时S3系鸡极显著高于与其他品系(P<0.01);在腿肌组织中,0周龄时F系极显著高于其他系(P<0.01),2、5周龄时S3系极显著高于F系和H系(P<0.01);在肝脏组织中,5、10、15周龄时F系鸡极显著高于S3系(P<0.01);在成肌细胞中,IMPAD1在增殖50%时的表达量显著高于分化期(P<0.05);在肌内脂肪细胞中,IMPAD1在分化7 d时的表达量显著低于增殖期与分化1、3、5 d(P<0.05)。相关性分析表明,IMPAD1与H系鸡的胸肌率呈极显著正相关(P<0.01)。综上所述,IMPAD1在不同品系和组织间的表达存在显著差异性,说明IMPAD1参与了不同品系鸡不同周龄的肌肉发育与脂肪沉积过程,该基因可作为相关性状筛选时的候选基因。
