摘要:
以海南省五指山野生桑黄子实体为原料,旨在通过科学手段优化其多糖成分的热水浸提工艺,并深入评估该多糖的体外抗氧化活性,以期为天然抗氧化剂的开发提供坚实的理论支撑。研究过程中,采用单因素试验与响应面分析法相结合的方式,系统探究提取温度(85、90、95 ℃)、提取时间(1.5、2.0、2.5 h)及料液比(1 ∶10、1 ∶20、1 ∶30)(g∶mL)这3个核心参数对多糖得率的具体影响,并采用ABTS+自由基清除法、羟自由基清除法以及铁离子还原能力法测定其抗氧化活性。结果表明,最佳提取工艺条件为温度85 ℃、时间2 h、料液比1 g ∶20 mL,在此条件下,多糖的实际得率高达0.71%,与模型预测值高度一致,充分验证了优化工艺的准确性。进一步分析显示,所得多糖总糖含量为4476%、还原糖含量为13.30%。尤为重要的是,体外抗氧化试验揭示,桑黄多糖对ABTS+自由基和羟自由基的清除能力展现出显著的浓度依赖性,最高清除率分别达93.29%和69.71%,接近阳性对照(维生素C)的效果;同时,其在 10 mg/mL 浓度时的铁离子还原力也达到了6.20 mmol/L。综上所述,本研究优化了海南省五指山野生桑黄多糖的提取工艺,明确其显著的体外抗氧化活性,为后续产业化开发奠定了基础。
