摘要:
采用RT-PCR与RACE技术相结合的方法,从山葡萄果皮中克隆到黄烷酮3-羟化酶(F3H)基因的cDNA全长序列。该基因全长1 398 bp,包含1 092 bp的完整开放阅读框,编码363个氨基酸,相对分子质量为40.81 ku,等电点(PI)值为5.37。二级结构预测表明,随机卷曲是VamF3H蛋白最大量的结构元件,不具有信号肽,属于不稳定亲水蛋白。氨基酸序列与欧亚种葡萄(FQ389950)、圆叶葡萄(KF040970)、美国蔓藤(JX087441)等的F3H类基因同源性较高,相似性分别为99%、99%、96%。
