摘要:
采用RT-PCR技术克隆家兔DGAT1部分基因片段,并采用半定量RT-PCR技术研究其在齐兴肉兔不同组织中的表达谱。研究结果获得了DGAT1基因960 bp的编码序列,共编码320个氨基酸。通过核苷酸和氨基酸序列比对分析发现,家兔DGAT1基因与其他物种的相似性在81.7%~89.2%之间;遗传距离分析表明,家兔与人的亲缘关系最近,与牛的最远。组织表达谱分析表明,DGAT1基因在心脏、肝脏、脾脏、肺脏、肾脏、胃、盲肠、淋巴结、背最长肌、皮下脂肪等多种组织中均有表达,且皮下脂肪中表达量最高,肺脏组织中表达量最低。研究结果为进一步研究DGAT1基因在家兔脂肪代谢中的生理功能提供依据。
