摘要:
DNA甲基化是一种重要的 DNA 修饰方式,它能在不改变 DNA一级结构的情况下调控基因的表达,能够维持正常的细胞功能,在胚胎发育、遗传印记中起重要作用。以黄鳝为对象,采用限制性内切酶-PCR检测特定DNA 片段是否发生甲基化的方法,研究不同温度、pH值、光照周期条件下DMRT基因在各组织中的甲基化差异。用甲基化敏感的限制性内切酶HpaⅡ酶切黄鳝的性腺、肾、脾、肝、肠等5个组织,根据已知的DMRT2、DMRT3、DMRT4 基因序列设计特异的引物扩增酶切产物,并设置未酶切对照组,用1%琼脂糖检测,分析差异条带,若未酶切组泳道出现条带,酶切组泳道也出现条带则判定为阳性结果,否则判定为阴性结果。结果表明:不同生态因子条件下,在5个组织中DMRT2-FR2扩增序列中的位点均表现为阳性结果;DMRT3-FR2、DMRT4-FR2、DMRT4-FR3扩增序列中的位点均表现为阴性结果,说明这4个序列中位点的甲基化状态非常稳定,环境因子的小幅变化对其影响不大。
