摘要:
为建立一种新的荧光原位杂交与流式细胞术相结合(FISH-FCM)的方法,以快速鉴定肉食品中的肠杆菌科细菌,采用与肠杆菌科细菌16S rRNA保守序列互补的探针Enter1432并验证其特异性。结果表明:6种肠杆科细菌与探针Enter1432杂交率均在90%以上,而5种非肠杆菌科细菌与探针杂交率均低于1%;与SN/T 0738—1997《出口食品中肠杆菌科检验方法》传统培养法相比,FISH-FCM定量检测法全程约需4~5 h,检测8份猪肉样品中肠杆菌科细菌数为1.4万~640万个/g,4份鸡肉样品中肠杆菌科细菌数为5.0万~21万个/g,检出率为100%。综上所述,研究建立的FISH-FCM法可快速检测肉品中的肠杆菌科细菌,适于食品卫生、临床实验室推广使用。
