摘要:
建立一种灵芝均一性菌种制作方法及其发酵工艺。超声破碎灵芝琼脂固体平板培养物,采用镜检和涂布平板法分析超声均质效果,以生物量和胞外多糖为指标优化其发酵工艺。结果表明:灵芝琼脂固体菌种破碎完全、均一性好,菌落数达到186.00 CFU/个(琼脂块直径0.5 cm);以超声均质化菌种为接种物于恒摇床发酵,生物量、胞外多糖分别为9.24、0.62 g/L,低于琼脂块菌种(分别为13.37、1.16 g/L);采用静置加摇床两阶段发酵方法(共10 d),以静置 3 d、摇床7 d为最优,生物量、胞外多糖含量分别达到15.14、0.86 g/L;与琼脂块菌种恒摇床发酵相比分别提高2996%、16.21%;超声均质菌种优化后的接种量为7 mL。
