摘要:
为研究1-脱氧野尻霉素(1-deoxynojirimycin,简称DNJ)对家蚕中肠BmSuc1基因表达及其酶活性的影响,以5龄第3天家蚕为试验材料,经口添食不同剂量的生物碱DNJ,采用实时荧光定量PCR方法检测中肠BmSuc1的表达情况,同时测定β-呋喃果糖苷酶的活性。结果表明,添食不同剂量的DNJ均能引起BmSuc1基因在添食6、9 h出现上调表达,但相对表达量有一定差异,添食4 μg/头的处理组在添食6、9 h基因的相对表达量分别是对照组的1.9、2.9倍,添食2 μg/头的处理组在添食6、9 h基因的相对表达量分别仅为对照组的1.5、1.8倍。另外,添食4 μg/头的处理组β-呋喃果糖苷酶活性在添食6、9、12 h显著高于对照组,而添食2 μg/头的处理组酶活性仅在添食6、9 h显著高于对照组。由结果可知,酶活性变化与基因的转录表达规律基本一致。
