摘要:
以玉米皮为原料,分析液料比、乙醇体积分数、提取温度和超声时间等单因素对类黄酮得率的影响,利用响应面法优化超声波辅助提取类黄酮物质工艺,优化玉米皮制备天然抗氧化剂的提取工艺;通过测定所提取类黄酮物质的清除自由基能力,评价玉米皮类黄酮物质的抗氧化活性。结果表明:玉米皮类黄酮物质提取的最佳工艺条件为料液比1 g ∶25 mL、乙醇体积分数50%、提取温度33 ℃、超声时间57 min,类黄酮物质的得率为12.96 mg/g;在优化条件下,类黄酮物质得率的试验验证值与预测值误差在±1.0%之内。在上述优化工艺前提下,发现玉米皮类黄酮物质对DPPH自由基、羟自由基、超氧阴离子自由基均具有较强清除作用,其半数抑制浓度分别为8.436、0.825、0.621 mg/mL。
