摘要:
研究建立同时测定珍珠粟直链和支链淀粉含量的检测方法,并通过测定分析13个种质资源中直链与支链淀粉含量的变化,为筛选适宜的加工品种提供依据。根据双波长选择原理和扫描光图谱,确定直链淀粉的测定波长、参比波长分别为634、426 nm,支链淀粉的测定波长、参比波长分别为539、763 nm。直链淀粉浓度在4~44 mg/L之间(r=0.999 3)、支链淀粉浓度在20~120 mg/L(r=0.999 3)之间,其碘复合物与吸光度呈良好的线性关系。结果表明,13个珍珠粟供试样品中直链、支链淀粉的含量分别为16.07%~23.48%、52.34%~73.80%,支链淀粉含量差异是引起总淀粉含量差异的主要因素。样品88、50、59、1-1的品种优良,可作为选种栽培、精深加工的优选品种。
