摘要:
采用DNA混池测序法筛选斑点叉尾[XCZ11.tif,SQ]肌肉生长抑制素(MSTN)基因SNP位点,同时使用SNaPshot法将筛选到的SNP多态性位点进行分型,最后与生长性状进行关联分析。共筛选到4个SNP位点,即g.3501 T>G、g.3844 T>A、g.4297 C>G、g.4355 G>C。其中,g.3501 T>G位于第一内含子;g.3844 T>A位于第二外显子,并造成密码子GUA变为GAA,氨基酸由缬氨酸(Val)变为谷氨酸(Glu);g.4297 C>G、g.4355 C>G位于第二内含子。关联分析结果表明,SNP位点g.4355 G>C与斑点叉尾[XCZ11.tif,SQ]的体质量和体长呈显著性负相关,具有GG基因型个体的体质量和体长显著性低于CC型和CG型(P<0.05),该位点可作为斑点叉尾[XCZ11.tif,SQ]分子育种的候选分子标记。
