摘要:
为探讨适合枸杞棉蚜基因组DNA提取的方法,采用9种方法提取枸杞棉蚜基因组DNA,并对每种方法提取的DNA样本质量进行综合比较分析。结果表明,改进十二烷基硫酸钠(SDS)法、优化KAc法、STE精提法、Chelex粗提法、十六烷基三甲基溴化铵(CTAB)法、盐析法均可从枸杞棉蚜中提取总DNA,浓度高且所制备出的DNA均可成功应用于mtDNA COⅠ基因扩增。STE粗提法、Chelex精提法、饱和NaCl法Ⅱ提取的枸杞棉蚜基因组DNA质量低,不能满足分析要求。改进SDS法及STE精提法所制备的DNA质量较佳、产量较高、实用性强,是值得推广应用的枸杞棉蚜基因组DNA提取方法。盐析法提取DNA,试剂成本低、毒性小,是一种安全有效的枸杞棉蚜基因组DNA提取方法。Chelex粗提法是一种快速的枸杞棉蚜基因组DNA提取方法。
