摘要:
采用低温液体富集、酪蛋白筛选平板、温度复筛等方法从黑龙江省大庆油田土壤这一非极端自然环境中筛选分离获得2株产低温蛋白酶的耐冷细菌LS3和LS4,通过菌落形态和显微观察,结合16S rDNA全序列分析确定前者为假单胞菌属(Pseudomonas),后者为微小杆菌属(Exiguobacterium)。LS3和LS4菌株在低温(10 ℃)下均能较好地生长及产酶,最适酶活温度分别为40、30 ℃,最适作用pH值均为8,对热敏感,低温仍能维持较好的酶活,属于低温碱性蛋白酶。其中,LS3菌株在含尿素培养基中产酶活性较高(52.46 U/mL),LS4菌株产的蛋白酶对盐有一定耐受性。本研究结果丰富了对低温蛋白酶的研究开发,为其在食品工业、农业复合菌剂等领域的应用提供了基础。
