摘要:
为了探讨稻草与黑麦草混合青贮发酵过程中微生物的动态变化规律,以稻草、黑麦草为青贮原料,从青贮后1、5、12、21、31、61、99、154 d的稻草、黑麦草混合青贮样中取样,分别测定pH值,采用培养方法计数乳酸菌、酵母菌、霉菌数量的变化,使用PCR-变性梯度凝胶电泳(PCR-DGGE)技术分析青贮过程中菌群组成多样性。结果表明:黑麦草与稻草混合青贮pH值在青贮初期下降迅速,乳酸菌数量在5d达到最高峰,然后逐渐下降,酵母菌数量呈先降后升再降等波动变化,稻草与黑麦草混合青贮过程中优势菌主要有植物乳杆菌(Lactoacillus plantarum)、类肠膜魏斯氏菌(Weissella paramesenteroides)、食窦魏斯氏菌(W. cibaria)、短乳杆菌(L.brevis),且随着青贮时间延长,优势菌数量下降。由结果可知,将DGGE和16S rRNA序列分析技术结合,能有效地研究青贮细菌的遗传多样性和组成的变化。
