摘要:
利用冷休克表达载体构建含有人胸腺肽α原(proTα)编码基因的质粒并在大肠杆菌单一蛋白生产(SPP)系统中表达单一的可溶性蛋白。以proTα mRNA基因序列为模板敲出ACA序列，设计并合成proTα-lessACA目的基因片段后，直接克隆到原核冷休克表达载体pColdⅡ(sp-4)上，将pColdⅡ(sp-4)-proTα-lessACA质粒转化到Escherichia coli BL21感受态细胞中，筛选阳性克隆后采用Nde Ⅰ和Hind Ⅲ限制
