摘要:
通过对猪乳汁Exosome中总miRNA进行分离提取,以验证其对猪繁殖与呼吸综合征病毒(PRRSV)的抑制作用。首先提取猪乳汁中的Exosome,并将提取的Exosome对感染PRRSV的MARC-145细胞进行初步处理,于24、48 h后进行病毒滴度测定,并在MARC-145细胞上接种PRRSV病毒96 h后对细胞形态进行观察;随后对猪乳Exosome中的总miRNA进行进一步提取,用提取的总miRNA处理感染PRRSV的Marc-145细胞,于24、48 h后进行病毒滴度测定,同时也在MARC-145细胞上接种PRRSV病毒96 h后对细胞形态进行观察。结果表明,转染猪乳Exosome和转染猪乳Exosome总miRNA细胞中的PRRSV滴度都显著低于转染PBS(阴性对照);同时相对于阴性对照组,转染猪乳Exosome和转染猪乳Exosome总miRNA的细胞形态排列较紧密,边缘较清晰整齐。说明猪乳Exosome中存在与抑制PRRSV复制有密切关系的miRNA,本研究将为PRRS的预防和治疗提供新的理论依据和途径。
