摘要:
家兔是研究基因功能和疾病模型的重要模式动物,但目前的兔遗传操作体系急需改进,建立成熟高效兔基因敲除平台具有重要意义。为了探究CRISPR/Cas9基因组编辑系统对家兔肌肉生长抑制素(myostatin,简称MSTN)基因的敲除效率,构建了Cas9/gRNA表达质粒pCas9/gRNA。将Cas9/gRNA质粒转染到兔纤维细胞,经DNA测序发现pCas9/gRNA诱导MSTN基因发生碱基缺失和点突变,基因突变效率为23%。结果表明,利用CRISPR/Cas9系统可在兔成纤维细胞中高效介导MSTN基因突变,为进一步建立高效的基因敲除兔平台奠定了基础。
