摘要:
通过对比聚酰胺、DM-130、LSA-40、DHP-600与FL-1等5种大孔树脂的静态“吸附-解吸”性能,筛选出最适合甜茶叶总黄酮分离纯化的大孔树脂,并对其动态纯化工艺条件进行探讨。结果表明,最适大孔树脂的动态“吸附-解吸”参数如下:上样液质量浓度为2.0 mg/mL,上样液体积为60 mL,上样液pH值为5.0,上样流速为 2 BV/h,洗脱剂乙醇体积分数为70%,洗脱剂用量为70 mL,洗脱流速为2 BV/h。在此优化条件下,得到甜茶叶总黄酮的纯度为39.6%。并且在抗氧化研究中,纯化后的甜茶叶总黄酮对2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)自由基的抗氧化能力半抑制浓度(IC50值)由108.7 μg/mL降低到77.6 μg/mL。由结果可知,甜茶叶总黄酮经过FL-1大孔树脂的纯化,能够提高抗氧化能力,因此,它具有很好的医药保健品生产前景。
