摘要:
旨在提出1种新的具有高灵敏度、特异性抗黄芪甲苷(AST)单克隆抗体的纯化方法,从而为制备具有类似活性基团的其他天然化合物的亲和层析树脂方法提供参考。用AST与环氧活化琼脂糖6B(EAS6B)偶联制备的免疫亲和柱从小鼠腹水中纯化抗AST的高敏单克隆抗体(mAbs),与用G-葡聚糖凝胶从小鼠腹水中纯化的抗AST单克隆抗体进行比较,通过间接ELISA(酶联免疫吸附测定)和间接竞争ELISA比较2种方法纯化的单克隆抗体,评价亲和培养基的效用。间接ELISA结果显示,用AST-EAS6B柱纯化的抗AST单克隆抗体的效价略高于用G-葡聚糖凝胶柱纯化的抗体效价;间接竞争ELISA结果显示,用AST-EAS6B柱纯化的抗AST单克隆抗体的结合抑制50%浓度(IC50)为 0.005 μg/mL,用G-葡聚糖凝胶柱纯化的抗AST单克隆抗体的IC50为0.05 μg/mL,用AST-EAS6B柱纯化的抗AST单克隆抗体比用G-葡聚糖凝胶柱纯化的抗AST单克隆抗体对AST更具有特异性。
