摘要:
为预测并鉴定猪丹毒丝菌甘油醛-3-磷酸脱氢酶(GAPDH)分子线性B细胞表位,首先通过多序列比分析已经全基因组测序的10株菌株的GAPDH一级结构保守性;然后使用在线软件Bepipred-2.0预测猪丹毒丝菌GAPDH线性B细胞表位,接下来通过人工化学合方式合成GAPDH表位对应的多肽;最后使用间接ELISA法检测这些多肽与猪丹毒丝菌GAPDH高免血清之间的反应。研究发现已经完成全基因组测序的10株菌株的GAPDH一级结构高度保守。通过软件预测,找到了猪丹毒丝菌GAPDH的3个潜在的表位。ELISA检测结果表明74VLSERDPKNLPWKELGV90178HAYTNDQNTLDGPHAKGDLRRGRAAAQSIIPNSTGA213 与GAPDH高免血清反应明显,是猪丹毒丝菌GAPDH的表位。成功预测并鉴定到2个猪丹毒丝菌的抗原表位,这些表位将为以后基于猪丹毒丝菌GAPDH表位的诊断、疫苗设计及致病机制研究提供技术基础。
